Fluorometric quantification of nucleic acids

Qubit® 2.0 Fluorometer (ThermoFisher Scientific) is a benchtop fluorometer for DNA, RNA and protein quantitation using the highly sensitive and accurate fluorescence-based specific probes.

Tecan Safire (Tecan) is a UV-Vis fluorescence and absorbance plate reader providing the high throughput and accuracy ideal for use in modern drug discovery and research. This microplate reader has high sensitivity, excellent performance and built in flexibility, in a compact format, that allows users to quickly change applications without cumbersome filter changes. With its fast wavelength scanning and high sensitivity, users get results in short measurement times.

Method of quantification :

PicoGreen dsDNA quantitation: selective quantification of dsDNA with a fluorescent DNA-binding dye from 0.2 to 100 ng/µl, with only 1 µl of DNA. This dye shows minimal binding to ssDNA and RNA, allowing specific quantitation of dsDNA.
RiboGreen RNA quantitation: selective quantification of ssRNA with a fluorescent RNA-binding dye from 1 to 500 ng/µl, with only 1 µl of RNA.

Absorbance quantification of nucleic acids

NanoVue plus spectrophotometer (GE Healthcare) is an UV/Visible spectrophotometer easy-to-use and reliable instrument for the quick and accurate quantitation of nucleic acid and protein samples.

Qualification by capillary electrophoresis of nucleic acids

Bioanalyzer 2100 system (Agilent Technologies) is a microfluidic platform that provides quantitation and quality control of DNA and RNA by capillary electrophoresis, providing high quality datas.
DNA analysis: The Bioanalyzer 2100 allows to separate, quantify and control your DNA fragments (PCR, fragmented or digested DNA, NGS libraries, etc.) with a high resolution (separate fragments down to 5 bp).
RNA analysis: With its RIN algorithm (RNA Integrity Number), the Bioanalyzer 2100 allows to control your total RNA sample integrity, also mRNA, depleted-rRNA and cRNA profiles.

QIAxcel system (Qiagen) fully automates the analysis of DNA fragments and RNA with sensitivity and high-resolution by replacing the traditional gel analysis by capillary micro-electrophoresis. The ready-to-run gel cartridges contain 12 separation micro-channels with a built-in gel matrix for fast, high-resolution analysis of up to 96 samples per run. QIAxcel cartridges can analyze all your PCR fragments, NGS libraries and others with a size range between 15 bp to 5 kb with a detection sensitivity of 0.1 ng/µl.
